We now wish to extend this work by studying how endothelial cells (which line the blood channels of the liver) and fibroblasts (which contribute to fibrosis) influence the migration of leucocytes into the liver and determine their fate in the diseased organ. To achieve this we will fluorescently tag the CLEVER-1 receptor and study its trafficking within human endothelial cells during leucocyte migration, and examine the contribution of fibroblasts to this process. We will probe the molecular mechanisms that drive this process and determine which soluble and cell-bound factors contribute to CLEVER-1-mediated injury. This project would require the use of immunohistochemistry, fluorescence microscopy, live cell imaging, plasmid DNA/siRNA manipulation, western blotting/ELISAs and cell culture techniques (including isolation and maintenance of primary human cells); all of which are routinely carried out in the Centre for Liver Research. In summary: we hypothesize that CLEVER-1 plays an important role in the leucocyte recruitment cascade, and propose that studies of the interplay between endothelial cells, fibroblasts and leucocytes will shed light on the therapeutic potential of CLEVER-1.
